Uridine is contraindicated in patients with:

  • Known allergy or hypersensitivity to uridine.
  • Pregnant and breastfeeding women.
  • Non-emergent treatment of side effects linked with fluorouracil or capecitabine, as it may reduce their efficacy.
  • Administration beyond 4 days or 96 hours following fluorouracil or capecitabine use.

Uridine triacetate is provided as granules in 10-gram packets for oral administration. In adults, the recommended dose is 1 packet every 6 hours for a total of 20 doses, with or without food. For children, the dose is 6.2 grams/m2 of body surface area every 6 hours for 20 doses, with or without food.

Here are the instructions for preparing and administering uridine granules:

  • Mix one packet of uridine granules (10 grams) with 4 ounces (100 ml) of soft food like applesauce, pudding, or yogurt in a small container.
  • Crush the contents of one full 10-gram packet into a fine powder and add it to 4 ounces (100 ml) of starch-based thickening product.
  • Maintain a ratio of no more than 1 gram per 10 ml of reconstituted food when giving to pediatric patients receiving less than 10 grams.
  • Administer the mixture within 30 minutes of mixing and ensure the patient swallows all the medicine with at least 4 ounces of water.
  • In children, adjust the dose based on their weight.
  • If vomiting occurs, administer another full dose as soon as possible, followed by the next dose at the regular scheduled time.
  • Measure the dose accurately using a scale or a graduated teaspoon.
  • Complete the full course of 20 doses as directed by the doctor.
  • If necessary, the medication can be given through a nasogastric tube or gastrostomy tube, particularly in cases of severe mouth inflammation (mucositis) or when the patient is in a coma.
